Aligning the Timing Belt Pulleys 200

I have a 1983 242 turbo (b21FT) that has given me a lot of trouble over the past few days. Currently I am working on timing, trying to align the pulleys for the crankshaft, camshaft, and the shaft that powers the distributor and waterpump. I have the notches in the pulley wheels aligned with the notches on the block for both the camshaft and crankshaft. However, I can't be too sure about the third shaft as the mark on the block is difficult to see from above. Is there any tricks I should know about when trying to align the pulleys? Also, is it possible for my brick to run if the camshaft and distributor shaft are misaligned by one tooth? I only ask this because it runs very rough (sometimes backfiring)even though all of the pulleys appear to be aligned.
