No, although it will effect the time it takes for it to get to that point. After about 20 minutes of driving, if you were to then check the ATF level at idle, in park, it should read at the full hot mark on the dipstick. Slightly lower (like in between low and full) shouldn't be a problem BUT it shouldn't be 1-2 quarts overfilled.

If one removes the trans drain plug, draining everything that'll fall out with gravity, you then put in slightly less than 4 qts to refill it (say 3 3/4qts).
