diagnostic computer question 200 1989

This is my first brick with a diagnostic computer, and I can't find the answer to this question anywhere: does it make a difference if the car is running or not when you run the diagnostic check? As I understand it, the computer stores up codes as it goes along until you clear them, but I can't figure out if it does anything different if you check for codes with the engine idling.
