I tuned my '74 progressively leaner until I could barely make it ping when it was very hot in a heavy load condition, a few degrees at a time.
But first, everything else should be set in stone, electrical and ignition quality and timing, fuel pressure, and vacuum leaks.
Get a t-handle 3mm allen wrench. The screw head gets filled with grit, so get a firm purchase. You can't adjust while running, really, it closes the air/gas flow/flap when you push on the screw and stalls the engine.
