Will it run at all if the timing marks are one tooth off?

Yes, the engine will run with the timing belt one tooth off. Like crap. BTDT

How crappy depends on whether the one tooth off is forwards or backwards, and whether the crank and cam timing is OK and the distributor gear is off, or the cam and dist are OK with each other and the crank is off, etc.

The fix is to do the job over again, sorry.
