The purpose of the canister is for fugitive vapor emissions. Rather than being released into the atmosphere they are stored in the carbon canister. (The granulated carbon in the canister temporarily stores the HydroCarbons), to be burned in the engine at the best possible time.
When the engine is under load (low vacuum but high flow), the engine is able to handle the stored up gasoline 'fumes' and the valve is opened (under the correct conditions by the ECM).
The 'fumes' enter the system post MAF so the ECM somehow compensates for the extra A/F, unless it is really a very small amount (dimimus) and not really an issue.
If the ECM does not have to compensate for this (because of the small volume), then plugging the line probably won't hurt. I don't know what would be gained either.
