I once drove 256 miles (most of that without overdrive) while pulling a U-Haul trailer (daughter moving into her freshman college dorm) over some hilly terrain (NJ to MA) -- locking out OD is strongly recommended when towing. That was in fall '99, almost nine years ago -- and I've since put almost 100K more miles on the car (still have it and use it daily) without transmission trouble, so it's safe to say there was no lasting damage.
I did get only get about 10mpg on the trip, but I can't say how much mileage loss was from staying in 3rd, and how much from pulling the trailer.
