First thing I would look for is a dividing clue. That is a clue that divides one system from another, e.g. the vacuum supply from the vacuum switching.
If the other buttons work, then I'd peel back the front of the console and probably expect to find a split hose at the defrost button. If they didn't, I'd take a look at the check valve under the hood and start tracing the missing vacuum supply back to the storage bottle behind the console panel.
